Barker was born in Fontana, California and began drumming when he was a kid. He started playing with The Aquabats in 1996, but left to join Blink-182 in 1998. Blink-182 encountered mainstream success through Enema of the State (1999). Barker made himself known as an incredibly versatile drummer, recording and playing in various music projects from a variety of music genres like hip hop, alternative rock, pop, and country. A reality TV series called Meet the Barkers starred Barker. The show was canceled after an accident on a plane in 2008, but returned and released his first solo album called Give the Drummer Some, in the year 2011. He continues to collaborate with rappers and released extended versions of his songs with Yelawolf Asher Roth, and Nottz, as well as with Blink-182 as well as the Transplants.Aside from drumming, he founded the clothing company Famous Stars and Straps in 1999. Then he founded the labels LaSalle Records in 2004 and DTA Records in 2019. Businesses like DC Shoes and Zildjian cymbals have co-designed products in his name. In the year 2015, he released his memoir titled Can I say: Living large in the world, cheating death and Drums Drums. Barker is also a vegan and invested in Crossroads Vegan Restaurant in Los Angeles.
